I'd like to get assistance with troubleshooting this error.
Generally, the program worked for 24-48 hours. Then I rebooted my computer. I may have also automatically did a registry cleaner that could have caused this issue, but I after giving the game a break, I came back to it and went to launch the game... and steam was not working. I identified (I believe) that the issue was that steam was being blocked my Norton Antivirus.
I did some troubleshooting on the STEAM client, which I resolved and my indepth troubleshooting has gotten me to the point that I've re-installed Steam and Rocksmith...
When I originally installed Rocksmith, I had the issue that I am now having, where .. after the profile selection, Rocksmith2014.exe would stop working. My earlier fix for this was an update of my video drivers. Unfortunately, now that I've updated them once, they don't need updated anymore!
I've created a long list of 'things' that i've done... of course.. this is a list that was put together after I had already been troubleshooting. I believe I have the majority of trial/error attempts cataloged and in order. But I might be missing something. I believe it is a directx driver issue.. but would appreciate help.
According to Steam's System Information Tab.. I'm running Win7-64bit on AuthenticAMD, 3314 Mhz, 6 processor with NVidia GeForce GTX 560, DirectX Driver Name: Atklumdisp.dll, Version 9.18.13.3221, DirectX Driver Version 7.14.10.304, Driver Date 19 Dec 2013, OpenGL Version 4.4
I find this difficult to believe.. being that the NVIDIA GEForce Experience is telling me my driver is the GeForce 332.21 driver that wasn't released until 1/7/2014..
What to do now?

I'm currently re-installing my Nvidia driver .. I'll re-post if this solves the issue. (But I don't want to retype this, if it doesn't!)
I thought I'd post what finally worked for me after days of trying to get this game to work in hopes of saving other people some frustration. Before I do, let me just say that I've never seen such a poorly managed software release. First we wait a year for the PC version, with new release dates set every two months, and then when it's finally released, it took me (and many others based on Google searches) days to get it running. The good news...I LOVE THIS GAME now that it's finally running.
Issue #1: Game doesn't launch. Steam runs fine, but when I would launch Rocksmith, I would get a brief 'launching' popup and then...nothing. No error message, no game, no nothing. I searched and searched and tried all of the following suggestions with no luck: uninstall and reinstall Steam, uninstall and reinstall Rocksmith, verify game cache, launch game from Rocksmith.exe directly, mess with Compatibility mode settings, run .exe as Administrator, created another Windows user account with nothing installed in it, disabled AVG antivirus, moved Rocksmith game folder to another directory, start in offline mode, update drivers, update steam client, install Steam on a different drive, run the directx executable directly, run the vc redist executable directly, and probably others I'm forgetting. NONE of these worked.
Solution: So what did work? For me, it was just setting the launch options to '-windowed'. Right-click on Rocksmith in your Steam client and go to Properties. Click the 'Set Launch Options' button. Type in -windowed. And now the game launches...in Full Screen mode...go figure. EDIT: I forgot I also have to uncheck the 'Enable Steam Community In-Game' option on the same Properties screen to get things to work.
Issue #2: Cable is unplugged. Um, no it's not. Guitar is plugged into cable, cable is plugged into computer. Come on...I finally solved Issue #1 (after days of trial and error) and now you don't recognize the plugged in guitar!? To the Google-mobile!
Solution: The answer to this one seems to be more straight-forward. You have to unplug any other USB devices (except for keyboard and mouse). This is a MAJOR pain in the rear for me as all of my PC's serve double duty as Windows Media Centers, so I have a USB-UART device plugged into every one of them in order to use an IR remote control and wireless 'air mouse' when watching TV/movies. But at least now I know. I have to crawl behind the computer, unplug my USB device, then plug in the guitar every time I want to play the game. Thanks Ubisoft!
Again, I think it's a testament to how great this game is that I am still happy even after dealing with these issues now that I can sit and play along while Eddie Vedder belts out Black. Great work Ubisoft! Now...you're all fired.
Hope this post helped somebody out there.
